Deafness Reversed in Weeks: Single Gene Therapy Injection Restores Hearing in Children and Adults
In a groundbreaking medical breakthrough, scientists have successfully restored hearing in both children and adults born with a rare genetic form of deafness—using just a single shot of gene therapy.
The results, published in Nature Medicine, reveal that all ten participants in the study began to experience hearing within weeks of receiving a viral injection carrying the healthy OTOF gene. For some, it marked the first time they had ever heard sound.
One of the most remarkable cases involved a seven-year-old girl who, just four months after treatment, was able to hold full conversations with her mother. Her rapid recovery was consistent with the overall study findings, where children aged five to eight showed the most significant improvements.

The therapy was developed by Otovia Therapeutics, in collaboration with institutions including Karolinska Institutet and several hospitals in China. It works by using a synthetic adeno-associated virus (AAV) to deliver a functional copy of the OTOF gene directly into the cochlea—the inner ear structure responsible for hearing. This gene encodes otoferlin, a vital protein needed to convert sound into electrical signals the brain can interpret.
“This is a major leap forward in treating genetic deafness, and we believe it’s only the beginning,” said Dr. Maoli Duan, a co-author of the study and consultant at Karolinska Institutet.
The trial involved ten patients aged between 1 and 24, all of whom had severe hearing impairment caused by mutations in the OTOF gene. The therapy was administered through a single injection into the round window of the cochlea. After one month, most participants had regained partial hearing. At six months, all ten showed substantial improvement, with average sound perception improving from 106 decibels to 52.
Children, particularly those between five and eight, demonstrated the fastest and most dramatic responses. Yet, the treatment also proved effective in teenagers and adults—marking the first time the technique has shown such broad success across age groups.
“Previous studies in China have shown promise in young children, but this is the first demonstration of strong hearing restoration in older patients as well,” said Duan. “We’re now closely monitoring all participants to assess how long the benefits last.”
Importantly, the gene therapy was well-tolerated. The only notable side effect was a temporary drop in neutrophils—a type of white blood cell—but no serious complications were observed over a 6 to 12-month follow-up.
Encouraged by this success, researchers are expanding their focus to more common forms of genetic deafness involving genes like GJB2 and TMC1. Although these present greater complexity, early tests in animal models have been promising.
The study was conducted with support from Zhongda Hospital, Southeast University, and multiple Chinese research programs, alongside Otovia Therapeutics Inc., which also employs many of the scientists involved.
